OR52K1 Gene

HGNC Family G protein-coupled receptors
Name olfactory receptor, family 52, subfamily K, member 1
Description Olfactory receptors interact with odorant molecules in the nose, to initiate a neuronal response that triggers the perception of a smell. The olfactory receptor proteins are members of a large family of G-protein-coupled receptors (GPCR) arising from single coding-exon genes. Olfactory receptors share a 7-transmembrane domain structure with many neurotransmitter and hormone receptors and are responsible for the recognition and G protein-mediated transduction of odorant signals. The olfactory receptor gene family is the largest in the genome. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nHuntingtin‐interacting protein 1 (HIP1) is a multifunctional endocytic adaptor that links clathrin‐mediated internalization to downstream signaling events in diverse cell types. In neurons, HIP1 interacts with mutant huntingtin and its binding partner Hippi to promote the assembly of a procaspase-8 recruitment complex that may trigger apoptosis in Huntington disease, while it also regulates the internalization of AMPA and NMDA receptors to fine-tune synaptic transmission and plasticity. In addition, HIP1’s overexpression is associated with oncogenic transformation through altered trafficking and stabilization of receptors such as the epidermal growth factor receptor and the transferrin receptor, highlighting its role in receptor turnover and cell survival."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its role in receptor trafficking, HIP1 interfaces with the cytoskeleton through direct binding to clathrin and actin, a dynamic interplay that is critical for maintaining cellular architecture and enabling proper vesicle formation. In neuronal presynaptic terminals, HIP1 modulates the abundance of key fusion proteins, influencing synaptic vesicle release and protecting against exacerbated dysfunction in the presence of toxic polyglutamine expansions. Moreover, HIP1 deficiency in murine models yields a spectrum of developmental abnormalities—including skeletal defects, impaired hematopoiesis, and compromised male reproductive capacity—underscoring its essential role in tissue maintenance and organ function."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nHIP1’s influence extends into immune regulation and oncogenic signaling. A homologous serine hydrolase encoded by Mycobacterium tuberculosis, also termed Hip1, subverts dendritic cell cytokine secretion and maturation, thereby altering antigen presentation and T-cell polarization. In cancer, HIP1 interacts with receptor tyrosine kinases such as c-Kit and EGFR, and its regulated phosphorylation plays a key role in receptor stabilization and maintenance of survival signals. Intriguingly, studies in murine models reveal that compensation—whether by expression of human HIP1 or through alternative splicing events that restore the open reading frame in mutant alleles—can rescue degenerative and tumorigenic phenotypes associated with HIP1 loss, further emphasizing the complexity and redundancy of its regulatory mechanisms."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "15"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "François G Gervais, Roshni Singaraja, Steven Xanthoudakis, et al.
