OR6C1 Gene

HGNC Family G protein-coupled receptors
Name olfactory receptor, family 6, subfamily C, member 1
Description Olfactory receptors interact with odorant molecules in the nose, to initiate a neuronal response that triggers the perception of a smell. The olfactory receptor proteins are members of a large family of G-protein-coupled receptors (GPCR) arising from single coding-exon genes. Olfactory receptors share a 7-transmembrane domain structure with many neurotransmitter and hormone receptors and are responsible for the recognition and G protein-mediated transduction of odorant signals. The olfactory receptor gene family is the largest in the genome. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAlthough the query mentioned OR6C1, the provided abstracts focus on the SCN11A‐encoded voltage‐gated sodium channel Nav1.9. Nav1.9 is predominantly expressed in small‐diameter nociceptive neurons where it mediates a persistent, tetrodotoxin‐resistant sodium current that amplifies subthreshold depolarizations. This amplification is critical for driving neuronal hyperexcitability under conditions of inflammation, thereby contributing to the generation and maintenance of inflammatory pain hypersensitivity. In various rodent models, deletion of Nav1.9 abolishes or reduces responses to pro‐inflammatory mediators without disrupting basal pain thresholds, confirming its role as an effector in peripheral sensitization and inflammatory hyperalgesia."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, Nav1.9’s contribution extends to diverse pain modalities. Gain‐of‐function mutations in SCN11A can lead to hyperexcitability in nociceptors, resulting in conditions such as cold‐aggravated pain, familial episodic pain, and even alterations in itch signaling. In contrast, loss‐of‐function studies (via genetic ablation or knock‐down) consistently show attenuated responses to inflammatory, visceral, or neuropathic stimuli. Several investigations have demonstrated that alterations in the channel’s gating properties (and its interaction with G-protein–coupled signaling pathways) affect the initiation and maintenance of spontaneous nerve activity, mediator release (e.g. from peptidergic terminals), and even conducted vasomotor responses in microcirculatory networks."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "21"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its well‐established role in pain and hyperalgesia, Nav1.9 also appears to influence functions in other sensory and non‐sensory systems. Evidence from studies in visceral afferents, the auditory pathway, and even in cells such as macrophages and retinal glia suggests that Nav1.9 activity is crucial for fine‐tuning excitability, mediating neurogenic inflammation, and shaping specialized action potentials in distinct cellular contexts. These additional roles hint at a broader importance of Nav1.9 in sensory processing and tissue homeostasis, and they underscore its potential as a high‐value target for therapeutic intervention in disorders ranging from migraine and inflammatory pain to certain forms of congenital insensitivity and pruritus."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "22", "end_ref": "27"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Birgit T Priest, Beth A Murphy, Jill A Lindia, et al.
