| HGNC Family | G protein-coupled receptors |
| Name | olfactory receptor, family 6, subfamily C, member 76 |
| Description | Olfactory receptors interact with odorant molecules in the nose, to initiate a neuronal response that triggers the perception of a smell. The olfactory receptor proteins are members of a large family of G-protein-coupled receptors (GPCR) arising from single coding-exon genes. Olfactory receptors share a 7-transmembrane domain structure with many neurotransmitter and hormone receptors and are responsible for the recognition and G protein-mediated transduction of odorant signals. The olfactory receptor gene family is the largest in the genome. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nLymphocyte activation gene‑3 (LAG‑3) is an inhibitory receptor expressed primarily on activated T cells—including conventional CD4⁺ and CD8⁺ T cells—as well as on regulatory T cells, activated B cells, and even certain dendritic cell subsets. Its engagement with MHC class II molecules (often in complex with specific peptides) results in negative regulation of T‐cell expansion, cytokine secretion, and effector function. In various experimental systems, LAG‑3 has been shown to restrain T‐cell proliferation and to calibrate the size of the memory and effector T‐cell pools, thereby contributing to self‐tolerance and dampening immune responses in settings such as autoimmunity and chronic infections."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAt the molecular level, LAG‑3 transduces its inhibitory signals via a distinct cytoplasmic domain that contains conserved motifs essential for binding downstream signaling molecules. Structural studies have revealed that LAG‑3, although related to the co‑receptor CD4, recognizes only a subset of stable peptide–MHC class II complexes and interacts with other ligands such as fibrinogen‑like protein 1 (FGL1). In addition, LAG‑3 is subject to proteolytic cleavage by metalloproteases (e.g., ADAM10 and ADAM17), a regulatory step necessary for full T‑cell activation. Moreover, LAG‑3 can physically associate with other inhibitory receptors like PD‑1, and this cooperation—mediated by overlapping signaling motifs—further reinforces inhibitory signals in T cells."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nPreclinical studies have underscored the therapeutic potential of modulating LAG‑3 activity. In diverse models—including autoimmunity, infectious diseases, and cancer—LAG‑3 blockade (often in combination with PD‑1 inhibition) has been shown to reinvigorate exhausted T cells, enhance effector functions, and promote tumor clearance. LAG‑3 deficiency or inhibition leads to altered cellular metabolism and enhanced downstream signaling (for example, via STAT5), rendering T cells less susceptible to regulatory cell–mediated suppression and more proficient at cytokine production. The broad impact of these regulatory effects has prompted exploration of LAG‑3 as a target for cancer immunotherapy as well as for interventions aimed at modulating autoimmunity, viral infections, and even inflammatory responses in the context of other diseases."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "34"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Creg J Workman, Kari J Dugger, Dario A A Vignali "}, {"type": "b", "children": [{"type": "t", "text": "Cutting edge: molecular analysis of the negative regulatory function of lymphocyte activation gene-3."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "J Immunol (2002)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.4049/jimmunol.169.10.5392"}], "href": "https://doi.org/10.4049/jimmunol.169.10.5392"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "12421911"}], "href": "https://pubmed.ncbi.nlm.nih.gov/12421911"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Creg J Workman, Dario A A Vignali "}, {"type": "b", "children": [{"type": "t", "text": "The CD4-related molecule, LAG-3 (CD223), regulates the expansion of activated T cells."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Eur J Immunol (2003)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1002/eji.200323382"}], "href": "https://doi.org/10.1002/eji.200323382"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "12672063"}], "href": "https://pubmed.ncbi.nlm.nih.gov/12672063"}]}, {"type": "r", "ref": 3, "children": [{"type": "t", "text": "Frédéric Triebel "}, {"type": "b", "children": [{"type": "t", "text": "LAG-3: a regulator of T-cell and DC responses and its use in therapeutic vaccination."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Trends Immunol (2003)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1016/j.it.2003.10.001"}], "href": "https://doi.org/10.1016/j.it.2003.10.001"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "14644131"}], "href": "https://pubmed.ncbi.nlm.nih.gov/14644131"}]}, {"type": "r", "ref": 4, "children": [{"type": "t", "text": "Creg J Workman, Linda S Cauley, In-Jeong Kim, et al.
