| Name | p21 (RAC1) activated kinase 5 |
| Description | The protein encoded by this gene is a member of the PAK family of Ser/Thr protein kinases. PAK family members are known to be effectors of Rac/Cdc42 GTPases, which have been implicated in the regulation of cytoskeletal dynamics, proliferation, and cell survival signaling. This kinase contains a CDC42/Rac1 interactive binding (CRIB) motif, and has been shown to bind CDC42 in the presence of GTP. This kinase is predominantly expressed in brain. It is capable of promoting neurite outgrowth, and thus may play a role in neurite development. This kinase is associated with microtubule networks and induces microtubule stabilization. The subcellular localization of this kinase is tightly regulated during cell cycle progression. Alternatively spliced transcript variants encoding the same protein have been described. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nP21‐activated kinase 5 (PAK5) is a member of the group B p21‐activated kinases that plays a pivotal role in regulating cytoskeletal dynamics and cellular homeostasis. Mechanistic studies have revealed that PAK5 negatively regulates kinases involved in microtubule destabilization; for instance, it inhibits MARK2 activity, thereby preventing the phosphorylation and detachment of tau protein from microtubules—a process implicated in early neurodegeneration. In addition, its proper subcellular trafficking—from mitochondria to the nucleus—is essential for its anti‐apoptotic function, as mislocalization abolishes its cell survival‐promoting effects."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn cancer biology, PAK5 has emerged as an important oncogenic factor. Elevated expression of PAK5 is observed in colorectal carcinoma, where it appears to decrease cell adhesion and promote migration and metastasis. In breast and gastric cancers, functional experiments indicate that PAK5 drives cell proliferation and invasion, partly through modulating the cell cycle and cytoskeletal reorganization. In addition, activating mutations in PAK5 have been linked to enhanced ERK pathway signaling in lung cancer, suggesting that aberrant PAK5 activity contributes to a spectrum of malignancies by reinforcing proliferative and migratory signals."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "3", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditional studies have further underscored the tumor‐promoting role of PAK5. Overexpression of the kinase has been noted in various tumor types, including osteosarcoma, where its depletion leads to reduced proliferation and enhanced apoptosis. Moreover, recent findings have uncovered that PAK5 can phosphorylate key substrates such as DDX5, thereby facilitating post‐translational modifications and microRNA processing (e.g. miR‑10b) that promote tumor cell metastasis. Collectively, these observations highlight PAK5 as a multifaceted regulator of cell survival, growth, and motility, making it an attractive therapeutic target for diverse human cancers."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "10"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Dorthe Matenia, Bettina Griesshaber, Xiao-yu Li, et al.
