HGNC Family | Proteoglycans |
Name | proline/arginine-rich end leucine-rich repeat protein |
Description | The protein encoded by this gene is a leucine-rich repeat protein present in connective tissue extracellular matrix. This protein functions as a molecule anchoring basement membranes to the underlying connective tissue. This protein has been shown to bind type I collagen to basement membranes and type II collagen to cartilage. It also binds the basement membrane heparan sulfate proteoglycan perlecan. This protein is suggested to be involved in the pathogenesis of Hutchinson-Gilford progeria (HGP), which is reported to lack the binding of collagen in basement membranes and cartilage. Alternatively spliced transcript variants encoding the same protein have been observed. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nPRELP is an extracellular matrix proteoglycan expressed in diverse connective tissues, notably cartilage and basement membranes. In inflammatory settings such as rheumatoid arthritis, fragments of PRELP are released into synovial fluid where they can modulate complement activation by binding key complement proteins (e.g., C9 and C3) to prevent membrane attack complex formation, thereby potentially dampening local inflammation and tissue damage. This complement‐inhibitory function highlights PRELP’s role as a local regulator of innate immunity in damaged tissues."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nPRELP also mediates cell adhesion and matrix organization. Its N-terminal domain, enriched in proline and arginine, interacts with cell surface glycosaminoglycans and specific proteoglycans (such as syndecan-1, syndecan-4, and glypican-1) to promote adhesion and focal adhesion formation, underscoring its importance in tissue integrity and remodeling. Moreover, differential tissue expression studies indicate that PRELP may function distinctively in connective tissues, affecting collagen fibril organization as demonstrated in transgenic mouse models."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "2", "end_ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of cancer, PRELP displays complex expression patterns and functional roles. Its expression is altered in several malignancies, including chronic lymphocytic leukemia, bladder cancer, and oral squamous cell carcinoma, where it may influence tumor behavior through modulation of epithelial–mesenchymal transition and cell adhesion. Notably, direct interactions with growth factor receptors like the insulin-like growth factor I receptor and p75NTR suggest that PRELP can regulate cellular proliferation and survival pathways. These cancer-related findings are complemented by observations in metastatic colorectal cancer where PRELP influences cellular biomechanics via integrin signaling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in inflammation and oncogenesis, PRELP contributes to host defense and tissue regeneration. It exhibits antibacterial properties in the respiratory tract by binding pathogens and enhancing complement-mediated killing, thereby supporting innate immunity against common respiratory bacteria. Additionally, PRELP expression is pivotal for ligament tissue reconstruction and may serve as a key inducer of mesenchymal stem cell differentiation into ligament-like tissue, a process that involves zinc-dependent signaling pathways. In parallel, altered PRELP levels in adipose tissue have been linked to obesity and adipose fibrosis via activation of the FAK/MAPK signaling cascade through its interaction with p75NTR, while an MMP-cleaved fragment of prolargin serves as a circulating biomarker in psoriatic arthritis.
