| HGNC Family | G protein-coupled receptors |
| Name | prostaglandin E receptor 4 (subtype EP4) |
| Description | The protein encoded by this gene is a member of the G-protein coupled receptor family. This protein is one of four receptors identified for prostaglandin E2 (PGE2). This receptor can activate T-cell factor signaling. It has been shown to mediate PGE2 induced expression of early growth response 1 (EGR1), regulate the level and stability of cyclooxygenase-2 mRNA, and lead to the phosphorylation of glycogen synthase kinase-3. Knockout studies in mice suggest that this receptor may be involved in the neonatal adaptation of circulatory system, osteoporosis, as well as initiation of skin immune responses. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nPTGER4, which encodes the prostaglandin E₂ receptor subtype EP4, plays a pivotal role in immune regulation and genetic susceptibility to inflammatory and autoimmune conditions. Genetic studies have linked altered PTGER4 expression to Crohn’s disease and aspirin‐intolerant asthma, underscoring its involvement in mucosal homeostasis. Moreover, EP4 signaling in dendritic cells facilitates their migration toward lymphoid chemokines, while in T‐helper cells it helps drive a proinflammatory Th17 phenotype through modulation of interleukin‐17 and interleukin‐10 expression. In macrophages, EP4 activity contributes to the attenuation of chemokine production via stabilization of NF‐κB regulators, and genome‐wide analyses in multiple sclerosis further hint at broader immunogenetic roles for PTGER4."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its immunomodulatory functions, EP4 signaling is critically involved in tumorigenesis and cancer progression across a spectrum of malignancies.
