| HGNC Family | Phosphatases |
| Name | protein tyrosine phosphatase, non-receptor type 18 (brain-derived) |
| Description | The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, the mitotic cycle, and oncogenic transformation. This PTP contains a PEST motif, which often serves as a protein-protein interaction domain, and may be related to protein intracellular half-live. This protein can differentially dephosphorylate autophosphorylated tyrosine kinases that are overexpressed in tumor tissues, and it appears to regulate HER2, a member of the epidermal growth factor receptor family of receptor tyrosine kinases. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAlthough the collection of studies provided does not directly address PTPN18, together they underscore the central importance of spatially and temporally regulated signaling cascades controlled by the Rho family GTPases—most notably Cdc42, Rac1, and RhoA—in a diverse array of cellular functions. These reports demonstrate that precise regulation of GTPase activation is essential for driving cytoskeletal reorganization, establishing cell polarity, coordinating membrane protrusions, and facilitating processes such as cell migration, neurite extension, exocytosis, stem cell maintenance, and tissue morphogenesis. For example, biosensor‐based approaches and conditional gene‐targeting have revealed that localized Cdc42 activation governs protrusion dynamics in migrating cells, the establishment of neuronal polarity and axon specification, and even the fine tuning of insulin granule release in pancreatic beta‐cells. Similar themes are extended to other systems such as podocyte foot process maintenance, dendritic spine plasticity, and vascular endothelial cell adhesion and lumen formation—all of which depend on the tight regulation of the actin cytoskeleton mediated by these GTPases."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "44"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast to the extensive mechanistic insights provided regarding Rho GTPase–driven signaling pathways, none of these abstracts mention or evaluate the function of the tyrosine phosphatase PTPN18. Although PTPN18 is known from other studies to function as a regulator of receptor signaling cascades through dephosphorylation events, its potential interplay or modulatory influence over the cellular processes highlighted here—such as cytoskeletal remodeling, cell adhesion, and migration—remains unexplored in the present literature. This disconnect highlights an important gap in our understanding and points to the need for future investigations to determine whether and how PTPN18 might interface with the dynamic networks governed by Rho GTPases to fine‐tune cellular behavior."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "44"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Gilles Gadéa, Laure Lapasset, Cécile Gauthier-Rouvière, et al.
