RAD54L Gene

Name RAD54-like (S. cerevisiae)
Description The protein encoded by this gene belongs to the DEAD-like helicase superfamily, and shares similarity with Saccharomyces cerevisiae Rad54, a protein known to be involved in the homologous recombination and repair of DNA. This protein has been shown to play a role in homologous recombination related repair of DNA double-strand breaks. The binding of this protein to double-strand DNA induces a DNA topological change, which is thought to facilitate homologous DNA paring, and stimulate DNA recombination. Alternative splicing results in multiple transcript variants encoding the same protein.[provided by RefSeq, Dec 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRAD54L is a Swi2/Snf2 ATP‐dependent DNA translocase that plays a central role in homologous recombination and DNA double‐strand break repair. It binds to Holliday junction–like structures with high specificity and promotes their bidirectional branch migration in an ATPase‐dependent manner. In doing so, RAD54L not only facilitates the extension of heteroduplex DNA during strand invasion but also acts in concert with RAD51—stimulating its DNA pairing activity and, once repair is initiated, promoting the dissociation of RAD51 from dsDNA. Furthermore, RAD54L’s ability to remodel chromatin, particularly by enabling RAD51 to invade nucleosomal templates, is governed by intricate mechanisms such as oligomerization and regulated ATP hydrolysis, with its activity modulated by distinct structural domains and phosphorylation events."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its biochemical functions, RAD54L has emerged as an influential factor in human disease, particularly in various cancers. Genetic variations and differential expression of RAD54L have been linked to clinical outcomes in pancreatic adenocarcinoma, where certain alleles correlate with reduced patient survival, and in other malignancies such as bladder cancer, lung adenocarcinoma, head and neck squamous cell carcinoma, choroid plexus carcinomas, and meningiomas. In these contexts, aberrant RAD54L function contributes to dysregulated cell cycle control, radioresistance, altered telomere dynamics, and enhanced oncogenic signaling via pathways including mTORC1; its expression is regulated by key transcription factors like E2F1 and Oct4. Moreover, while screening in familial chronic lymphocytic leukemia did not reveal a strong predisposition linked to RAD54L, studies in mesenchymal stem cells and models of alternative telomere lengthening indicate that proper RAD54L activity is critical for maintaining genomic stability. Intriguingly, recent evidence also suggests that modulating RAD54L expression may have therapeutic implications in non‐malignant conditions such as osteoarthritis, where overexpression can attenuate disease progression through downregulation of hypoxia-inducible and angiogenic factors."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "22"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Dmitry V Bugreev, Olga M Mazina, Alexander V Mazin "}, {"type": "b", "children": [{"type": "t", "text": "Rad54 protein promotes branch migration of Holliday junctions."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Nature (2006)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1038/nature04889"}], "href": "https://doi.org/10.1038/nature04889"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "16862129"}], "href": "https://pubmed.ncbi.nlm.nih.gov/16862129"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "Zhaoqing Zhang, Hua-Ying Fan, Joseph A Goldman, et al.
