Biochemical studies have shown that RHPN2 binds both GDP- and GTP-bound RhoA, thereby contributing to the disassembly of actin stress fibers and regulating cell morphology. In glioblastoma, RHPN2 gene amplification has been implicated in driving a mesenchymal transformation through activation of RhoA, resulting in an invasive phenotype that correlates with poor clinical outcomes. Similarly, enhanced RHPN2 expression in ovarian and lung adenocarcinomas promotes cell proliferation, migration, and invasion by engaging integrin β1/NF‑κB signaling and the RhoA/ROCK pathway. In thyrocytes, the related p76RBE protein is upregulated in response to cAMP signaling, where its interactions with RhoB and cytoskeletal components may influence intracellular trafficking and secretion.
