Name | ribosomal protein S6 kinase, 70kDa, polypeptide 1 |
Description | This gene encodes a member of the ribosomal S6 kinase family of serine/threonine kinases. The encoded protein responds to mTOR (mammalian target of rapamycin) signaling to promote protein synthesis, cell growth, and cell proliferation. Activity of this gene has been associated with human cancer. Alternatively spliced transcript variants have been observed. The use of alternative translation start sites results in isoforms with longer or shorter N-termini which may differ in their subcellular localizations. There are two pseudogenes for this gene on chromosome 17. [provided by RefSeq, Jan 2013] |

It is activated downstream of the mTOR complex and, when active, phosphorylates multiple substrates involved in translation initiation. For example, inactive RPS6KB1 is tethered to the eukaryotic initiation factor 3 (eIF3) complex, and upon mTOR‐dependent phosphorylation it dissociates to phosphorylate targets such as PDCD4 and eIF4B, thereby relieving translational repression and enhancing protein production. Moreover, factors deposited on spliced mRNAs recruit activated S6K1 to boost the pioneer round of translation. Through its kinase activity, it contributes to the phosphorylation of mTOR itself as well as upstream regulators, thereby attenuating insulin receptor substrate function and fine‐tuning growth factor responses. By phosphorylating key serine residues on IRS‐1 and even components of mTOR complex 2 (such as Rictor), S6K1 establishes feedback loops that modulate insulin sensitivity and oncogenic signaling. In breast, ovarian, esophageal, and other cancers, overexpressed or constitutively active S6K1 phosphorylates substrates such as estrogen receptor α and Gli1 to enhance proliferation, invasion, and angiogenesis.
