RUBCN Gene

Name rubicon autophagy regulator
Description The protein encoded by this gene is a negative regulator of autophagy and endocytic trafficking and controls endosome maturation. This protein contains two conserved domains, an N-terminal RUN domain and a C-terminal DUF4206 domain. The RUN domain is involved in Ras-like GTPase signaling, and the DUF4206 domain contains a diacylglycerol (DAG) binding-like motif. Mutation in this gene results in deletion of the DAG binding-like motif and causes a recessive ataxia. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Apr 2014]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRubicon, the protein product of KIAA0226, is a multifunctional regulator of intracellular membrane trafficking that plays a central role in autophagy and endocytic maturation. It is an integral subunit of the Beclin‑1–Vps34/class III phosphatidylinositol-3-kinase complex, where it negatively regulates autophagosome maturation by interfering with PI3KC3 lipid kinase activity. Rubicon associates with distinct Beclin‑1 complexes that include either Atg14L or UVRAG, with mutually exclusive binding partners modulating early autophagosome formation versus later maturation steps. In addition, Rubicon directly interacts with Rab7 via its RUN domain—as further corroborated by structural studies and its homolog PLEKHM1—to regulate endosome maturation and lysosomal trafficking, a process further reinforced by its role in LC3‑associated phagocytosis (a noncanonical autophagy pathway)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its role in autophagy, Rubicon functions as a key modulator of innate immune responses. It interacts with the p22^phox subunit of the NADPH oxidase complex to promote reactive oxygen species production during phagocytosis and inflammatory cytokine release, while also serving as a feedback inhibitor of proinflammatory signaling complexes (e.g. the CARD9–BCL10–MALT1 scaffold). Moreover, Rubicon dampens virus‐induced type‑I interferon responses by binding to key regulatory factors such as NEMO, thereby facilitating viral replication in infections such as hepatitis B and C. These immune‐regulatory functions underscore Rubicon’s dual capacity to both restrain and promote signaling events, a balance that is critical for the proper progression of the endocytic pathway and host defense mechanisms."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "14"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nClinically, dysregulation or mutation of Rubicon has been implicated in diverse human diseases. Frameshift mutations in KIAA0226 that disrupt the C‑terminal diacylglycerol binding‐like motif lead to mislocalization of Rubicon and are associated with recessive cerebellar ataxias, highlighting its role in vesicular trafficking. Moreover, epigenetic modifications involving the Rubicon gene have been linked to the progression and survival of non‑small‑cell lung cancer, while in the context of neurodegeneration, altered neuronal Rubicon activity appears to relieve its repressive effect on APP/amyloid‑β secretion, potentially exacerbating Alzheimer’s disease pathology."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "15", "end_ref": "19"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Kohichi Matsunaga, Tatsuya Saitoh, Keisuke Tabata, et al.
