SEPTIN9 Gene

Name septin 9
Description This gene is a member of the septin family involved in cytokinesis and cell cycle control. This gene is a candidate for the ovarian tumor suppressor gene. Mutations in this gene cause hereditary neuralgic amyotrophy, also known as neuritis with brachial predilection. A chromosomal translocation involving this gene on chromosome 17 and the MLL gene on chromosome 11 results in acute myelomonocytic leukemia. Multiple alternatively spliced transcript variants encoding different isoforms have been described.[provided by RefSeq, Mar 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nSEPTIN9’s aberrant methylation in circulating cell‐free DNA has emerged as a robust, minimally invasive biomarker for several solid tumors—most notably colorectal cancer—as well as for cancers of the head and neck, liver, lung and breast. Multiple independent case–control studies have validated that hypermethylation of the SEPTIN9 promoter correlates strongly with tumor occurrence and progression, thereby supporting its use in early detection, screening and even recurrence monitoring. In addition, detailed analyses have revealed complex, region‐specific methylation patterns and isoform expression changes that may further refine its prognostic and diagnostic power."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAt the cellular level, SEPTIN9 is a key member of the septin family orchestrating cytoskeletal organization and successful cell division. It localizes with actin filaments, microtubules and the mitotic spindle—playing an instrumental role during cytokinesis, especially at the final abscission step where specific isoforms mediate the recruitment of vesicle‐tethering complexes. SEPTIN9 forms hetero‐oligomeric complexes with other septins (such as SEPT2, SEPT7, SEPT11 and others) that are essential for proper filament assembly and for maintaining cell polarity. Detailed immunocytochemical and biochemical studies have characterized isoform‐specific localizations and interactions that underscore its necessity in coordinating actin–microtubule dynamics during mitosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "32"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its structural roles, SEPTIN9 actively modulates key signaling pathways that impact oncogenic processes as well as non‐dividing cellular functions. Specific isoforms—notably SEPTIN9_v1—bind and stabilize regulatory proteins such as HIF‑1α and components of the MAP kinase cascade (including JNK), promoting transcriptional programs that drive proliferation, angiogenesis and metastasis. In parallel, mutations in SEPTIN9 have been linked to hereditary neuralgic amyotrophy, underscoring its influence on neural cell integrity. Additional studies have implicated SEPTIN9 in receptor trafficking, mechanotransduction in endothelial cells, and even in orchestrating host–pathogen interactions by organizing cytoskeletal cages around intracellular inclusions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "33", "end_ref": "48"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Michael Scott, Paula L Hyland, Gordon McGregor, et al.
