| Name | small EDRK-rich factor 1B (centromeric) |
| Description | This gene is part of a 500 kb inverted duplication on chromosome 5q13. This duplicated region contains at least four genes and repetitive elements which make it prone to rearrangements and deletions. The repetitiveness and complexity of the sequence have also caused difficulty in determining the organization of this genomic region. This gene is the centromeric copy which is identical to the telomeric copy. Alternatively spliced transcripts have been documented but it is unclear whether alternative splicing occurs for both the centromeric and telomeric copies of the gene. The gene encodes a protein of unknown function which bears low-level homology with the RNA-binding domain of matrin-cyclophilin, a protein which colocalizes with small nuclear ribonucleoproteins (snRNPs) and the SMN1 gene product. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nStudies on the SERF protein family have revealed that these small, largely unstructured proteins act as evolutionarily conserved modifiers of protein aggregation. Initial work in Caenorhabditis elegans identified MOAG‐4 as a positive regulator of the formation of compact polyglutamine aggregation intermediates, and subsequent work showed that its human orthologs—including SERF1 isoforms—promote protein aggregation and related cytotoxicity in models of amyloid diseases. Such findings support a role for SERF proteins in accelerating the formation of fibrillar aggregates that are characteristic of multiple age‐related neurodegenerative disorders."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBiophysical and biochemical analyses have further defined the mechanism by which SERF proteins function. In vitro studies demonstrated that SERF1a, for example, can specifically distinguish between amyloid and nonamyloid aggregation pathways by engaging in early and transient interactions with amyloidogenic proteins, thereby provoking a marked increase in the formation of soluble amyloid nucleation templates. This activity accelerates fibrillization processes as seen with proteins such as α‑synuclein and amyloid‑β, where SERF1a binding promotes alterations in secondary structure and aggregation kinetics that ultimately enhance cytotoxicity."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, complementary studies have shown that the aggregation-promoting activity of SERF proteins depends on specific molecular interactions. The accumulation of negatively charged and hydrophobic, aromatic amino acid residues in aggregation-prone protein segments appears to mediate binding with SERF, triggering structural changes that lower the colloidal stability of the protein solution and enhance primary nucleation. These insights collectively illustrate that SERF proteins operate as dedicated facilitators of early amyloidogenic events, likely contributing to the proteotoxic signatures observed in neurodegenerative diseases."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Tjakko J van Ham, Mats A Holmberg, Annemieke T van der Goot, et al.
