TAFA4 Gene

Name TAFA chemokine like family member 4
Description This gene is a member of the TAFA family which is composed of five highly homologous genes that encode small secreted proteins. These proteins contain conserved cysteine residues at fixed positions, and are distantly related to MIP-1alpha, a member of the CC-chemokine family. The TAFA proteins are predominantly expressed in specific regions of the brain, and are postulated to function as brain-specific chemokines or neurokines, that act as regulators of immune and nervous cells. Alternatively spliced transcript variants have been observed for this gene. [provided by RefSeq, Nov 2011]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nTAFA4, also known as FAM19A4, is a secreted chemokine‐like molecule whose promoter methylation has been extensively studied for its diagnostic application in cervical cancer screening. In a series of studies, aberrant hypermethylation of the TAFA4/FAM19A4 gene in cervical scrapes or self‐samples of high‐risk HPV–positive women has emerged as a robust molecular biomarker to detect advanced cervical intraepithelial neoplasia (CIN2/3 and CIN3+) and cervical cancer. The marker has been validated in diverse cohorts and sample types, showing high sensitivity for detecting cervical carcinoma and advanced dysplastic lesions while providing increased specificity compared to conventional cytology or HPV16/18 genotyping. These findings support its clinical utility for triaging HPV‐positive women and aiding in the early and accurate identification of premalignant and malignant cervical lesions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its value as a molecular marker, TAFA4 functions as a multifunctional cytokine with a clear role in the regulation of immune responses. Experiments in monocytes and macrophages have shown that TAFA4 is upregulated upon lipopolysaccharide stimulation and in M1‐polarized cells, where it exerts chemotactic effects, enhances phagocytosis through increased Akt phosphorylation, and acts as a novel ligand for the formyl peptide receptor 1 (FPR1). Moreover, in the context of allergic inflammation, TAFA4 modulates mast cell activity by transcriptionally downregulating the expression of the high‐affinity IgE receptor (FcεRI) via a TAFA4–PTEN–PU.1 signaling axis. This downregulation results in attenuated antigen‐induced mast cell activation and reduced allergic responses, highlighting TAFA4’s potential as a therapeutic target in anti‐allergy interventions."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "17"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Lise M A De Strooper, Chris J L M Meijer, Johannes Berkhof, et al.
