In ESCC tissues, both TMEM161B-AS1 and its downstream target HIF1AN are notably downregulated compared to normal samples, with their reduced expression being significantly associated with advanced TNM stage, lymph node metastasis, and poor patient prognosis. Functionally, forced overexpression of TMEM161B-AS1 or inhibition of miR‑23a‑3p suppresses ESCC cell proliferation, invasion, and glycolytic metabolism—evidenced by diminished glucose consumption and lactate production. Mechanistically, TMEM161B-AS1 acts as a competitive endogenous RNA by sponging miR‑23a‑3p, thereby enhancing HIF1AN expression and contributing to the inhibition of tumor-promoting phenotypes.
