| HGNC Family | Ion channels, Protein phosphatase 1 regulatory subunits (PPP1R) |
| Name | transient receptor potential cation channel, subfamily C, member 5 |
| Description | This gene belongs to the transient receptor family. It encodes one of the seven mammalian TRPC (transient receptor potential channel) proteins. The encoded protein is a multi-pass membrane protein and is thought to form a receptor-activated non-selective calcium permeant cation channel. The protein is active alone or as a heteromultimeric assembly with TRPC1, TRPC3, and TRPC4. It also interacts with multiple proteins including calmodulin, CABP1, enkurin, Na(+)-H+ exchange regulatory factor (NHERF ), interferon-induced GTP-binding protein (MX1), ring finger protein 24 (RNF24), and SEC14 domain and spectrin repeat-containing protein 1 (SESTD1). Fundamental biophysical studies have shown that TRPC5 activation is initiated by G protein–coupled receptor stimulation and further modulated by intracellular Ca²⁺ elevations, specific lipid messengers such as lysophosphatidylcholine, and phosphoinositides. Its gating properties are finely tuned by redox signals, pH changes, and critical protein–protein interactions – for example through binding partners like calmodulin and SESTD1 – that couple receptor activation to downstream Ca²⁺ entry without necessarily altering its membrane density. Detailed patch‐clamp and optogenetic experiments have delineated its intrinsic properties, including modulation of open probability and sensitivity to store depletion."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "15"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the nervous system, TRPC5 is expressed in both central and peripheral neurons, where it contributes to sensory transduction and neuronal plasticity. In cardiac tissue, enhanced TRPC5 expression has been observed in failing hearts, where it may contribute to maladaptive hypertrophy via Ca²⁺/calcineurin–NFAT signaling. In inflammatory diseases such as rheumatoid arthritis, extracellular reduced thioredoxin activates TRPC5 in synoviocytes, influencing secretory and inflammatory responses. In several cancers – including breast, colorectal, and glioma – elevated TRPC5 levels, and its packaging into extracellular vesicles, promote chemoresistance by triggering downstream pathways that up‐regulate multidrug efflux transporters, stimulate autophagy, and even reprogram metabolism, while also affecting cell differentiation and migration.
