UGT1A1 Gene

HGNC Family Glycosyltransferases
Name UDP glucuronosyltransferase 1 family, polypeptide A1
Description This gene encodes a UDP-glucuronosyltransferase, an enzyme of the glucuronidation pathway that transforms small lipophilic molecules, such as steroids, bilirubin, hormones, and drugs, into water-soluble, excretable metabolites. This gene is part of a complex locus that encodes several UDP-glucuronosyltransferases. The locus includes thirteen unique alternate first exons followed by four common exons. Four of the alternate first exons are considered pseudogenes. Each of the remaining nine 5' exons may be spliced to the four common exons, resulting in nine proteins with different N-termini and identical C-termini. Each first exon encodes the substrate binding site, and is regulated by its own promoter. The preferred substrate of this enzyme is bilirubin, although it also has moderate activity with simple phenols, flavones, and C18 steroids. Mutations in this gene result in Crigler-Najjar syndromes types I and II and in Gilbert syndrome. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nUGT1A1 is a critical phase‐II conjugating enzyme that converts lipophilic compounds into water‐soluble metabolites through glucuronidation. In cancer chemotherapy, its most prominent function is the detoxification of SN-38, the active metabolite of irinotecan. By catalyzing SN-38 glucuronidation, UGT1A1 reduces drug bioavailability and toxicity – a property that underlies observations of severe diarrhea and neutropenia in patients receiving irinotecan. Numerous studies have documented that common UGT1A1 polymorphisms (including *28, *6, and other coding or promoter variants) markedly influence enzyme activity, leading to interindividual differences in irinotecan clearance and thus affecting both the occurrence of adverse events as well as the potential for dose escalation in selected patients."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn parallel to its role in drug metabolism, UGT1A1 is indispensable for the conjugation and clearance of endogenous substrates—most notably bilirubin. Reduced UGT1A1 activity resulting from genetic variants such as UGT1A1*28 contributes to conditions like Gilbert’s syndrome and may underlie altered cardiovascular risk profiles and liver function abnormalities. Beyond bilirubin, UGT1A1 also participates in the metabolism of estrogens and other bioactive molecules, while its expression is regulated by xenobiotic receptors and oxidative stress–responsive elements. Collectively, these attributes underscore the enzyme’s dual importance in both xenobiotic detoxification and physiological homeostasis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "34"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Y Ando, H Saka, M Ando, et al.
