| Name | UDP glucuronosyltransferase 1 family, polypeptide A8 |
| Description | This gene encodes a UDP-glucuronosyltransferase, an enzyme of the glucuronidation pathway that transforms small lipophilic molecules, such as steroids, bilirubin, hormones, and drugs, into water-soluble, excretable metabolites. This gene is part of a complex locus that encodes several UDP-glucuronosyltransferases. The locus includes thirteen unique alternate first exons followed by four common exons. Four of the alternate first exons are considered pseudogenes. Each of the remaining nine 5' exons may be spliced to the four common exons, resulting in nine proteins with different N-termini and identical C-termini. Each first exon encodes the substrate binding site, and is regulated by its own promoter. The enzyme encoded by this gene has glucuronidase activity with many substrates including coumarins, phenols, anthraquinones, flavones, and some opioids. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nUGT1A8 is an extrahepatic UDP‐glucuronosyltransferase that plays a critical role in the detoxification of xenobiotics and endogenous compounds. It is predominantly expressed in the gastrointestinal tract—with inducible expression also observed in liver cells—and its promoter architecture (involving Sp1/initiator-like elements) helps to dictate its tissue‐specific transcription. Multiple studies have identified common polymorphisms (for example, the A173G substitution and the *2/*3 alleles) at the UGT1A8 gene locus, which contribute to significant interindividual variation in enzyme expression and activity. These genetic variations have been explored with respect to their potential impact on drug metabolism and detoxification of environmental carcinogens."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFunctionally, UGT1A8 mediates the glucuronidation of a broad spectrum of substrates. It has been shown to inactivate carcinogenic 4‐hydroxy‐catechol estrogens and to conjugate active metabolites of drugs such as tamoxifen, mycophenolic acid, prunetin, and raloxifene. In vitro studies using recombinant UGT isoforms have demonstrated that certain UGT1A8 variants—for example, those harboring amino acid substitutions (e.g. the 173Ala/277Tyr variant)—exhibit markedly reduced or even undetectable glucuronidation activity toward these substrates. Such isoform‐specific differences highlight UGT1A8’s importance in influencing both the metabolic clearance and the pharmacological or toxicological outcomes of its substrates."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nClinically, the polymorphic behavior of UGT1A8 has been associated with altered drug pharmacokinetics and differential risk for adverse events as well as carcinogenesis. For instance, in renal transplant recipients the presence of UGT1A8 polymorphisms has been correlated with variations in mycophenolate mofetil metabolism and may contribute to gastrointestinal side effects; similarly, in carcinogenesis studies, specific UGT1A8 activity genotypes have been linked to a modified risk of esophageal and colorectal cancer.
