| HGNC Family | Glycosyltransferases |
| Name | UDP glucuronosyltransferase 1 family, polypeptide A9 |
| Description | This gene encodes a UDP-glucuronosyltransferase, an enzyme of the glucuronidation pathway that transforms small lipophilic molecules, such as steroids, bilirubin, hormones, and drugs, into water-soluble, excretable metabolites. This gene is part of a complex locus that encodes several UDP-glucuronosyltransferases. The locus includes thirteen unique alternate first exons followed by four common exons. Four of the alternate first exons are considered pseudogenes. Each of the remaining nine 5' exons may be spliced to the four common exons, resulting in nine proteins with different N-termini and identical C-termini. Each first exon encodes the substrate binding site, and is regulated by its own promoter. The enzyme encoded by this gene is active on phenols. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nUDP‐glucuronosyltransferase 1A9 (UGT1A9) is a liver‐enriched enzyme that plays a central role in catalyzing glucuronidation reactions to detoxify a broad spectrum of endogenous compounds and xenobiotics. It is instrumental in the inactivation and clearance of clinically relevant compounds, such as the active metabolite SN‑38 of irinotecan, mycophenolic acid, and even carcinogenic catecholestrogens. Molecular characterization studies have identified promoter variants—including the UGT1A9*22 allele with a thymidine insertion that enhances transcription—and demonstrated that expression of UGT1A9 is under the control of liver‐specific transcription factors such as HNF4α. These findings underscore its pivotal role in hepatic drug metabolism and detoxification."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFunctional investigations have revealed that specific genetic polymorphisms of UGT1A9—including promoter single‐nucleotide polymorphisms (such as –275T>A and –2152C>T) and amino acid substitutions (for example, M33T and C3Y)—can significantly modify its catalytic activity and substrate affinity. Such changes have been shown to influence the pharmacokinetics of drugs like irinotecan and mycophenolic acid—impacting both therapeutic efficacy and toxicity profiles—as demonstrated in various patient cohorts and integrated pharmacogenetic models. These studies collectively highlight the enzyme’s importance as a genetic determinant in personalized medicine strategies, accounting for interindividual variability in drug metabolism."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "6", "end_ref": "15"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, UGT1A9’s substrate versatility extends to extrahepatic processes, such as ethanol glucuronidation, where it demonstrates the highest intrinsic clearance among UGT isoforms—further emphasizing its broad role in xenobiotic metabolism.
