Description This gene encodes a member of the WIPI or SVP1 family of WD40 repeat-containing proteins. The protein contains seven WD40 repeats that are thought to fold into a beta-propeller structure that mediates protein-protein interactions, and a conserved motif for interaction with phospholipids. Analysis of individuals with homozygous pathogenic variants in WDR45B revealed a distinct neurodevelopmental syndrome characterized by profound developmental delay, early‐onset refractory epilepsy, progressive spastic quadriplegia with contractures, and marked structural brain abnormalities including ventriculomegaly, reduced cerebral white matter, and thinning of cerebral gray matter. These clinical observations underscore the essential role of WDR45B in neurodevelopment and suggest that disruptions in its function can lead to severe intellectual disability and global developmental delay.
