XIST Gene

HGNC Family Non-coding RNAs
Name X inactive specific transcript (non-protein coding)
Description X inactivation is an early developmental process in mammalian females that transcriptionally silences one of the pair of X chromosomes, thus providing dosage equivalence between males and females. The process is regulated by several factors, including a region of chromosome X called the X inactivation center (XIC). The XIC comprises several non-coding and protein-coding genes, and this gene was the first non-coding gene identified within the XIC. This gene is expressed exclusively from the XIC of the inactive X chromosome, and is essential for the initiation and spread of X-inactivation. The transcript is a spliced RNA. Alternatively spliced transcript variants have been identified, but their full length sequences have not been determined. Mutations in the XIST promoter cause familial skewed X inactivation. Structural domains—including its evolutionarily conserved A‐repeat region—and post‑transcriptional modifications such as N6‑methyladenosine (m6A) deposition (mediated by RBM15/15B and METTL3, and recognized by YTHDC1) are critical for establishing and maintaining this silenced state. Mechanistically, XIST is often found to function as a competing endogenous RNA (ceRNA) that sponges specific microRNAs (such as miR‑152, miR‑92b, miR‑200b‑3p, miR‑34a, miR‑21‑5p, among others) thereby modulating the expression of key oncogenes or tumor suppressors (for example, KLF2, SMAD7, E2F3, PDCD4) and influencing critical signaling pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "30"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its roles in epigenetic silencing and tumorigenesis, XIST participates in a broader network of cellular processes. XIST also influences immune cell function and has been implicated in non‑cancer pathologies such as myocardial injury, osteoarthritis, and acute kidney injury.
