{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n AP5S1 is a subunit of the AP‐5 adaptor protein complex, an evolutionarily ancient machinery that plays a critical role in endosomal sorting and lysosomal homeostasis. The AP‐5 complex functions in the transport of specific cargo molecules from late endosomal compartments, a process that is essential for maintaining proper lysosomal function and membrane trafficking. Disruption of AP‐5 complex components, including AP5S1, has been linked to the accumulation of membrane‐surrounded vesicular deposits and altered lysosomal enzyme profiles, phenomena that are thought to contribute to neurodegenerative conditions such as hereditary spastic paraplegia (HSP) (1,2,3,4). In neurons—which are particularly dependent on efficient endolysosomal processing—the failure of proper AP‐5 function can lead to axonal degeneration and the progressive loss of motor neurons and Purkinje cells. Moreover, the functional integration of AP‐5 with other HSP‐associated proteins (such as SPG11 and SPG15) underscores its relevance not only in cargo sorting but also in broader cellular processes, including selective autophagy and possibly even aspects of DNA repair.
