Description This gene encodes a transmembrane glycoprotein related to the type I receptors of the transforming growth factor-beta (TGF-beta) family, whose members play important roles in signal transduction in many developmental and pathological processes. The encoded protein however is a pseudoreceptor, lacking an intracellular serine/threonine kinase domain required for signaling. Similar proteins in frog, mouse and zebrafish function as negative regulators of TGF-beta, which has led to the suggestion that the encoded protein may function to limit the signaling range of the TGF-beta family during early embryogenesis. It functions primarily as a modulator at the crossroads of TGF‑β family and Wnt/β‑catenin signaling. Several studies in diverse pathophysiological settings have shown that BAMBI acts as a negative regulator of TGF‑β, activin, and bone morphogenetic protein (BMP) signals by interfering with receptor complex formation and by cooperating with inhibitory molecules such as Smad7 (3,4,8). In certain cancers—including colorectal and hepatocellular carcinomas—BAMBI is transcriptionally induced by Wnt/β‑catenin signaling, and its aberrant up‐regulation contributes to tumor progression by inhibiting TGF‑β–mediated growth arrest while enhancing cell motility and metastasis (2,3,6,7,9). In the fibrotic liver, toll-like receptor–mediated activation of NF‑κB results in BAMBI down‑regulation in hepatic stellate cells, which in turn potentiates TGF‑β signaling and promotes fibrogenesis (8). In diabetic retinopathy, a reduction in BAMBI owing to microRNA regulation further amplifies TGF‑β activity and mediates retinal vascular dysfunction (5). Conversely, in the pressure‑overloaded myocardium, increased BAMBI expression appears to serve as a protective feedback mechanism that limits excessive TGF‑β–dependent remodeling (10). In the central nervous system, decreased BAMBI levels have been associated with enhanced TGF‑β signaling that modulates pain perception via the endogenous opioid system (1,12). Moreover, in endothelial cells, BAMBI negatively regulates alternative TGF‑β cascades—thereby influencing capillary growth and vascular homeostasis (13)—and in ovarian granulosa cells, its expression is modulated by luteinizing hormone to affect folliculogenesis and luteinization (14). Finally, in pulmonary tissues, up‑regulation of BAMBI in response to infection contributes to an imbalance in TGF‑β signaling that may participate in tissue remodeling associated with chronic obstructive pulmonary disease (15).
