Description Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid. They show extensive diversity in tissue distribution and in their subcellular localization. CA XIV is predicted to be a type I membrane protein and shares highest sequence similarity with the other transmembrane CA isoform, CA XII; however, they have different patterns of tissue-specific expression and thus may play different physiologic roles. Structural and biochemical studies have revealed that CA XIV adopts an α‐CA fold similar to other mammalian isozymes, but with unique features that enable the design of selective inhibitors (1,6). In the central nervous system, CA XIV contributes to extracellular buffering and works jointly with CA IV to mitigate pH transients during synaptic transmission, although CA IV appears to be more critical in the hippocampus (2). In the retina, CA XIV is expressed in Müller glial cells and the retinal pigment epithelium, and its absence leads to significant deficits in light responses, indicating an important role in maintaining the proper ionic microenvironment necessary for normal visual function (3). In skeletal muscle, CA XIV is localized both in the sarcolemmal and sarcoplasmic reticulum membranes, accounting for a substantial fraction of the membrane-associated CA activity; however, genetic ablation studies indicate that loss of CA XIV is largely compensated and does not markedly impair contractile performance (5). Finally, the emerging potency of novel sulfonamide derivatives against CA XIV—as demonstrated by crystallographic analyses and inhibition studies—underscores its potential as a drug target for conditions such as certain retinopathies, epilepsy, and cancer (4).
