| Description | This antimicrobial gene belongs to the subfamily of small cytokine CC genes. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene displays chemotactic activity for dendritic cells, thymocytes, and activated macrophages but is inactive on peripheral blood lymphocytes and neutrophils. In the gut, CCL25 is produced by intestinal epithelial and endothelial cells and binds selectively to CCR9 expressed on effector/memory T cells and IgA antibody–secreting cells. This interaction is essential for imprinting gut‐homing properties onto activated lymphocytes, as dendritic cells in Peyer’s patches induce the up‐regulation of CCR9 on T cells so that they can respond to CCL25 and traffic to the small intestine (1,2,3,4). In addition, blockade or neutralization of CCL25 has been shown to reduce the recruitment of CCR9+ cells into the intestinal lamina propria, thereby diminishing mucosal IgA responses (2,3). In the thymus, CCL25/CCR9 signaling contributes to T cell development by mediating the attraction of developing thymocytes; indeed, TECK functions as a chemoattractant for immature T cells, an activity confirmed by its identification as the ligand for the previously orphan receptor later designated CCR9 (4,5). Moreover, in disease states such as necrotizing enterocolitis, increased TLR4‐dependent up‐regulation of the CCR9/CCL25 axis facilitates lymphocyte influx into the intestine, which can exacerbate local inflammatory responses (6).
