{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n CD72 is a key inhibitory coreceptor expressed on B cells that plays a crucial role in fine‐tuning B cell receptor (BCR) signaling and maintaining immunological tolerance. By virtue of its immunoreceptor tyrosine‐based inhibitory motifs, CD72 recruits phosphatases (such as SHP‐1) to dampen BCR‐mediated activation, thereby preventing overactivation and autoimmunity. Engagement of its natural ligand CD100 (also known as Sema4D) disrupts this inhibitory complex, transiently releasing the brakes on B cell activation so that appropriate immune responses can be mounted. Alterations in this CD72–CD100 regulatory circuit have been implicated in disease settings; for instance, diminished CD72 expression in neoplastic B cells contributes to enhanced survival and proliferation in B cell chronic lymphocytic leukemia, and dysregulation of CD72 signaling is associated with the breakdown of peripheral tolerance in autoimmunity. In addition, CD72 participates in the cross‐talk between regulatory B cells (Bregs) and regulatory T cells (Tregs), and its expression may serve as a diagnostic marker in settings such as minimal residual disease in B‐cell precursor acute lymphoblastic leukemia.
