Description This gene encodes a cell division cycle protein with kinase activity that is critical for the G1/S transition. The yeast homolog is also essential for initiation of DNA replication as cell division occurs. Overexpression of this gene product may be associated with neoplastic transformation for some tumors. CDC7 phosphorylates specific subunits of the MCM replicative helicase—most notably MCM2, MCM4, and MCM6—which promotes the subsequent recruitment of Cdc45 onto chromatin and thereby triggers the activation (“firing”) of replication origins (1,2,3,4,5,9). In addition, checkpoint signals elicited by DNA damage or replication stress can inhibit CDC7 activity to prevent unscheduled origin activation (1,9). Aberrant CDC7 activity is observed in many tumor types, where overexpression—often in conjunction with increased levels of its regulatory partner Dbf4—correlates with compromised p53 function, and inhibition of CDC7 in cancer cells leads to replicative failure and p53‐independent apoptotic cell death, suggesting its potential as a therapeutic target (6,7,8). Furthermore, recent genetic association studies have identified variants near the CDC7 locus that correlate with optic disc area and glaucoma, indicating that the functional repertoire of CDC7 may extend into ocular development and disease (10).
