Description This gene encodes a member of the type II transmembrane serine protease class of the trypsin superfamily. Members of this family are composed of multiple structurally distinct domains. The encoded protein converts pro-atrial natriuretic peptide to biologically active atrial natriuretic peptide, a cardiac hormone that regulates blood volume and pressure. This protein may also function as a pro-brain-type natriuretic peptide convertase. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n Corin is a cardiac‐expressed type II transmembrane serine protease that plays a central role in cardiovascular homeostasis by converting the inactive pro–atrial natriuretic peptide (pro–ANP) into bioactive ANP, a hormone critical for natriuresis, vasodilation, and blood pressure regulation (5,2,14). Its catalytic activity depends on precise zymogen activation, regulated by proteolytic cleavage and post‐translational modifications, including N‐glycosylation and ectodomain shedding mediated by ADAM10, which serve to spatially restrict its activity and prevent excessive proteolysis (8,10,18). In addition, corin activation is facilitated by upstream convertases such as PCSK6, underscoring the multistep regulation required for proper natriuretic peptide processing (9). Genetic variants that impair corin’s zymogen activation—for example, the T555I/Q568P allele and mutations such as R539C—have been associated with deficient natriuretic peptide processing, salt‐sensitive hypertension, and cardiac hypertrophy, suggesting that corin deficiency contributes to the pathogenesis of hypertensive heart disease and heart failure (4,6,7,13,16,17). Beyond its classical role in the heart, corin is also expressed in the pregnant uterus where it promotes trophoblast invasion and uterine spiral artery remodeling, and reduced uterine corin expression has been linked to pre‐eclampsia (1). Moreover, altered corin expression has been implicated in kidney disorders associated with salt retention, signifying its broader role in sodium homeostasis (20). The promoter and domain organization of the corin gene provide further insight into its strict tissue‐specific expression and regulation, while the detection of soluble corin in plasma highlights its potential utility as a biomarker for heart failure and other cardiovascular disorders (5,14,15,19).
