Description This gene encodes the heavy chain subunit of axonemal dynein, a large multi-subunit molecular motor. Axonemal dynein attaches to microtubules and hydrolyzes ATP to mediate the movement of cilia and flagella. The gene expresses at least two transcript variants; additional variants have been described, but their full length nature has not been determined. [provided by RefSeq, Jul 2008]

Studies indicate that DNAH9 localizes primarily to the distal compartment of the ciliary axoneme, where it is essential for the structural integrity and proper assembly of type 2 ODAs. Its function is crucial for generating the coordinated ciliary bending required for efficient mucociliary clearance and the establishment of left–right body asymmetry; loss‐of‐function mutations in DNAH9 lead to subtle yet clinically significant impairments in distal ciliary motility and are associated with laterality defects and forms of primary ciliary dyskinesia (1,2). In the male reproductive system, DNAH9 is also an integral component of the sperm flagellum’s ODA composition, with mutations contributing to nonsyndromic severe asthenozoospermia due to compromised sperm tail stability and motility (3). Moreover, genetic studies have implicated variants in the DNAH9 gene in modulating respiratory responsiveness, potentially through gene–environment interactions that influence bronchial hyperresponsiveness (4).
