Name DNA (cytosine-5-)-methyltransferase 3-like
Description CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. This gene encodes a nuclear protein with similarity to DNA methyltransferases, but is not thought to function as a DNA methyltransferase as it does not contain the amino acid residues necessary for methyltransferase activity. However, it does stimulate de novo methylation by DNA cytosine methyltransferase 3 alpha and is thought to be required for the establishment of maternal genomic imprints. This protein also mediates transcriptional repression through interaction with histone deacetylase 1. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2012]

Although it lacks intrinsic methyltransferase activity, DNMT3L functions as an adaptor and stimulatory factor by binding directly to the catalytic domains of DNMT3A and DNMT3B. This interaction enhances their enzymatic efficiency by promoting conformational changes that increase coenzyme (S‐adenosylmethionine) binding and facilitate efficient DNA methylation. Moreover, DNMT3L specifically recognizes histone H3 tails that are unmethylated at lysine 4 (H3K4me0) via its unique domains, thereby targeting its partner methyltransferases to appropriate genomic loci. This histone “reading” function is crucial for directing de novo methylation at imprinting control regions and dispersed repetitive elements, as well as for the proper establishment of maternal and paternal methylation imprints during gametogenesis. In male germ cells, DNMT3L is essential for the genome‐wide methylation of retrotransposons and other dispersed repeats, ensuring genome stability and normal meiotic progression. In embryonic stem cells, DNMT3L not only stimulates methylation of gene bodies of housekeeping genes but—through interaction with the Polycomb repressive complex—it can also counteract de novo methylation at promoters of bivalent developmental genes, thereby contributing to the proper epigenetic balance during cell differentiation. DNMT3L additionally cooperates with other chromatin regulators, such as CTCFL and PRMT7, to orchestrate the complex epigenetic events necessary for imprint establishment.
