It functions by interacting with key cytoskeletal components—most notably microtubules and their associated proteins such as MAP2 and βIII‐tubulin—to regulate neuronal migration, axonal guidance, dendritic outgrowth, and synapse formation. Functional analyses have shown that specific missense mutations in DPYSL5 impair its ability to form the ternary complex with MAP2 and βIII‐tubulin, thereby disrupting normal dendritic development. This disruption contributes to brain malformations (including corpus callosum agenesis and posterior fossa abnormalities) and is associated with variable degrees of intellectual disability. In addition, studies indicate that DPYSL5 can modulate neurite outgrowth by antagonizing tubulin polymerization—thereby acting as a negative regulator under certain developmental contexts—and thus may counterbalance the promoting effects of other CRMP family members.
