Transient expression of ETV2 is essential to commit multipotent mesodermal precursors to both endothelial and hematopoietic cell fates while simultaneously suppressing alternative lineages such as cardiac muscle. ETV2 directly induces key downstream regulators—including SCL/TAL1, GATA2, FLI1, and LMO2—to establish a gene regulatory network that drives vasculogenesis and hematopoiesis. In addition, ETV2 is a potent reprogramming factor that alone is capable of converting human somatic cells (for example, fibroblasts) into functional vascular endothelial cells, highlighting its potential for regenerative applications. Its expression is tightly modulated by extracellular signals such as VEGF and the cAMP/PKA–CREB pathway, which activate ETV2 transcription at critical stages of differentiation. Importantly, maintaining a transient, rather than constitutive, expression of ETV2 is necessary; prolonged expression perturbs normal vascular and blood development by forcing an extended endothelial program at the expense of hematopoiesis. Once the initial program is established, ETV2 activates other ETS family members (for example, FLI1) that subsequently help to sustain and mature the endothelial lineage.
