It functions primarily as a binding partner of FANCM, helping to target and stabilize the FA core complex at sites of stalled replication forks and DNA interstrand crosslinks. Its intrinsic DNA‐binding activity enables FAAP24 to recognize and bind to DNA structures that mimic repair intermediates, thereby facilitating the proper recruitment of repair proteins. In particular, FAAP24 is essential for achieving normal FANCD2 monoubiquitination—a critical modification for FA pathway activation—and for the efficient assembly of repair factors, including the recruitment of replication protein A (RPA) at DNA damage sites. This role not only supports the assembly of a repair-competent complex but also promotes ATR/Chk1‐mediated checkpoint signaling.
