Early studies showed that endogenous FAM170B becomes enriched first in round spermatids and then relocates adjacent to the Golgi and acrosomal region during sperm maturation. In this context, FAM170B interacts with the Golgi‐associated protein GOPC, suggesting it participates in acrosome formation and the acrosome reaction—a critical exocytotic event required for fertilization. Notably, while in vitro inhibition of FAM170B can block the acrosome reaction, in vivo studies using knockout mouse models revealed that absence of FAM170B does not adversely affect sperm count, motility, morphology, or overall fertility.
