Initial characterization showed that FAM3D is differentially expressed in a tissue‐specific manner (e.g. in the placenta) and is secreted following a classical secretory pathway (1). Functionally, FAM3D acts as a potent chemokine‐like mediator by binding with high affinity to the formyl peptide receptors FPR1 and FPR2, which facilitates the chemoattraction of neutrophils and monocytes (2,3). In vascular pathology, FAM3D engagement of these receptors can trigger intracellular signaling cascades (involving Gi‐protein and β‐arrestin pathways) that promote Mac‐1–mediated neutrophil recruitment, thereby exacerbating inflammatory conditions such as abdominal aortic aneurysm (2). In the gastrointestinal tract, where FAM3D is constitutively expressed, its secretion is regulated by nutritional status, suggesting a role in maintaining mucosal homeostasis and modulating inflammatory responses; in fact, both its upregulation during colitis and its deficiency—associated with impaired mucosal integrity, dysregulated epithelial proliferation, and enhanced sensitivity to colitis‐related tumorigenesis—underscore its importance in gut barrier function (3,4,9). In addition, emerging evidence highlights the potential of FAM3D as a biomarker in colorectal malignancy, as altered methylation patterns and expression levels contribute to prognostic models and may aid in the detection of circulating tumor cells (5,7).
