| Description | The protein encoded by this gene is a member of the interleukin 1 cytokine family. The activity of this cytokine is mediated by interleukin 1 receptor-like 2 (IL1RL2/IL1R-rp2), and is specifically inhibited by interleukin 1 family, member 5 (IL1F5/IL-1 delta). Interferon-gamma, tumor necrosis factor-alpha and interleukin 1, beta (IL1B) are reported to stimulate the expression of this cytokine in keratinocytes. The expression of this cytokine in keratinocytes can also be induced by a contact hypersensitivity reaction or herpes simplex virus infection. It is produced by epithelial cells—including keratinocytes, lung epithelium, intestinal stromal cells, and cervicovaginal epithelia—in response to microbial products, physical injury, or cytokine stimulation. Once released, IL‐36γ signals via a receptor complex composed of IL‐1Rrp2 and IL‐1RAcP to activate NF‐κB and MAPK pathways, thereby amplifying the production of inflammatory mediators such as TNF‐α, IL‐6, IL‐8, and chemokines like CCL20. In the skin, elevated IL‐36γ expression correlates with psoriatic lesions and functions as both a specific biomarker and an amplifier of IL‐17 and TNF–driven inflammatory circuits. Similar roles have been noted in mucosal and pulmonary tissues, where IL‐36γ is induced by infectious and danger signals to promote antimicrobial defenses through the induction of type 1 cytokines and licensing of macrophage polarization. Furthermore, recent studies indicate that IL‐36γ can be harnessed in a therapeutic context—for example, engineered CAR T cells secreting IL‐36γ exhibit improved expansion, persistence, and antitumor efficacy, while its expression in colonic myofibroblasts and in the cervicovaginal compartment suggests roles in both local host defense and inflammation.
