Name interleukin 36 receptor antagonist
Description The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine was shown to specifically inhibit the activation of NF-kappaB induced by interleukin 1 family, member 6 (IL1F6). This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. Two alternatively spliced transcript variants encoding the same protein have been reported. In normal physiology, IL‑36Ra binds to the IL‑36 receptor and prevents the recruitment of the IL‑1 receptor accessory protein, thereby antagonizing the activities of proinflammatory IL‑36 agonists (IL‑36α, IL‑36β, and IL‑36γ). Disruptive mutations in IL36RN result in a loss of function of IL‑36Ra, which leads to unopposed IL‑36 signaling, enhanced induction of downstream proinflammatory mediators, and increased production of neutrophil chemokines. These molecular events are associated with the development of autoinflammatory skin disorders such as generalized pustular psoriasis (GPP). In patients harboring recessive IL36RN mutations, the absence or inadequacy of functional IL‑36Ra correlates with early onset and severe pustular phenotypes distinct from chronic plaque psoriasis (1,2,3,4,5). Moreover, studies have shown that proper post‐translational processing (for example, via cleavage by neutrophil elastase) is required to generate the active form of IL‑36Ra that efficiently counteracts IL‑36–driven inflammation (6).
