| HGNC Family | Ion channels |
| Name | potassium channel, inwardly rectifying subfamily J, member 10 |
| Description | This gene encodes a member of the inward rectifier-type potassium channel family, characterized by having a greater tendency to allow potassium to flow into, rather than out of, a cell. The encoded protein may form a heterodimer with another potassium channel protein and may be responsible for the potassium buffering action of glial cells in the brain. Mutations in this gene have been associated with seizure susceptibility of common idiopathic generalized epilepsy syndromes. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nKCNJ10 encodes the inwardly rectifying potassium channel Kir4.1, a protein that is central to maintaining extracellular K⁺ homeostasis in the central nervous system. In astrocytes and other glial cells, Kir4.1 facilitates rapid K⁺ uptake following neuronal repolarization, thereby helping to stabilize the resting membrane potential and modulate neuronal excitability. Numerous genetic and animal‐model studies have demonstrated that abnormal Kir4.1 function—whether by loss‐of‐function mutations or gain‐of‐function alterations—can lead to impaired K⁺ buffering, delayed K⁺ clearance, and aberrant neuronal bursting. Such dysfunction has been strongly implicated in a spectrum of neurological disorders including epilepsy, ataxia, and even mood disturbances."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the kidney, Kir4.1 is predominantly localized to the basolateral membrane of cells in the distal convoluted tubule, where it plays an indispensable role in K⁺ recycling. This recycling is necessary for the proper operation of the Na⁺/K⁺-ATPase and thus for effective NaCl reabsorption and electrolyte balance. Loss-of-function mutations in KCNJ10 are a central cause of the autosomal recessive EAST (Epilepsy, Ataxia, Sensorineural deafness, and Tubulopathy) or SeSAME syndrome, a disorder characterized by renal salt wasting (resulting in hypokalemia, hypomagnesemia, and metabolic alkalosis). Functional investigations have revealed that altered pH sensitivity and trafficking defects in mutant Kir4.1 channels disrupt basolateral K⁺ conductance and membrane polarization, thereby impairing renal tubular transport."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "21", "end_ref": "28"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its roles in the brain and kidney, Kir4.1 is also expressed in the inner ear—most notably in cochlear supporting cells—where it contributes to the generation and maintenance of the endocochlear potential required for normal auditory transduction. Genetic studies have linked mutations in KCNJ10 to nonsyndromic hearing loss and inner ear malformations, sometimes acting in concert with other genes such as SLC26A4. Furthermore, emerging evidence suggests that aberrant immune responses against Kir4.1 may be important in the pathogenesis of demyelinating disorders, underscoring its broad importance in sensory and neural physiology."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "29", "end_ref": "34"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "R J Buono, F W Lohoff, T Sander, et al.
