NAA10 Gene

HGNC Family N(alpha)-acetyltransferase subunits (NAA)
Name N(alpha)-acetyltransferase 10, NatA catalytic subunit
Description N-alpha-acetylation is among the most common post-translational protein modifications in eukaryotic cells. This process involves the transfer of an acetyl group from acetyl-coenzyme A to the alpha-amino group on a nascent polypeptide and is essential for normal cell function. This gene encodes an N-terminal acetyltransferase that functions as the catalytic subunit of the major amino-terminal acetyltransferase A complex. Mutations in this gene are the cause of Ogden syndrome. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Jan 2012]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nNAA10, also known as ARD1, is the catalytic core of the NatA complex responsible for the co‐translational N‑terminal acetylation of a large fraction of eukaryotic proteins. Through its association with auxiliary subunits (such as NAA15/NATH) and binding partners like HYPK, NAA10 helps determine substrate specificity and regulates protein stability, folding, and interactions. Moreover, its activity is not strictly limited to co‐translational events; studies have shown that isolated NAA10 may also exhibit post‑translational acetylation—for example, modifying actin N‑termini and participating in noncanonical modifications such as N‑terminal propionylation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nSeveral laboratories have examined the interplay between NAA10 and hypoxia‐inducible factor‐1α (HIF‑1α). Some studies indicate that NAA10 directly binds HIF‑1α and, via acetylation, can enhance its interaction with ubiquitin ligases—thereby promoting degradation under normoxic conditions—while other reports find that modulation of NAA10 expression does not necessarily alter HIF‑1α stability. In addition, NAA10 interaction with HIF‑1α appears to influence other signaling pathways such as Wnt, suggesting that oxygen availability and even post‐translational modifications (for example, oxygen‐dependent hydroxylation events) might re‐tune its substrate preferences."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "14"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMutations in NAA10 have been directly linked to a spectrum of X‑linked developmental disorders.
