Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nNKp44, encoded by the NCR2 gene, is an activating receptor with a unique structural organization that distinguishes it from other immunoglobulin‐like molecules. Detailed structural analyses have shown that its extracellular Ig‐like domain forms a distinctive dimeric “saddle‐shaped” arrangement and that its surface expression relies on noncovalent association with the signaling adaptor DAP12 rather than on intrinsic inhibitory motifs. Moreover, alternative splicing of NCR2 gives rise to distinct NKp44 isoforms that may have different functional outcomes, underscoring the importance of this receptor in tailoring immune responses."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nA wide array of studies has demonstrated that NKp44 plays multifaceted roles in immune surveillance against pathogens and tumors. In the context of infection, NKp44 directly recognizes viral envelope proteins—including those from influenza viruses and various flaviviruses—as well as components of other pathogens, thereby triggering NK cell degranulation and cytokine production. In parallel, NKp44 expressed on natural killer as well as innate lymphoid cell subsets contributes to tissue homeostasis and inflammatory disorders; for example, shifts in NKp44⁺ ILC3 frequencies and cytokine‐secretion profiles have been linked to psoriasis and altered immune responses in settings as diverse as hepatitis E infection and xenotransplantation. Moreover, NKp44 engagement by tumor ligands—such as a cell‐surface isoform of proliferating cell nuclear antigen (PCNA) or an alternatively spliced isoform of MLL5 (NKp44L)—can either stimulate cytotoxicity against cancer cells or, paradoxically, result in inhibitory signals, NK cell apoptosis, or even immune evasion by tumors. These findings collectively illustrate that NKp44 interactions—with ligands expressed on transformed cells, virally infected targets, or even normal stromal and chondrocyte cells—are central to modulating NK cell effector functions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "6", "end_ref": "32"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its well‐documented roles in target cell recognition, emerging evidence highlights the regulatory versatility of NKp44 through differential splicing and context‐dependent ligand engagement. Distinct splice variants of NCR2 have been associated with divergent prognostic outcomes in acute myeloid leukemia and other malignancies, while tissue‐specific expression patterns—in placental and decidual cells or even on subsets of CD8⁺ T cells—suggest that NKp44 may sculpt adaptive immune responses, limit hyper-expansion of effector clones, and modulate cytokine‐driven inflammation. Furthermore, therapeutic strategies aimed at disrupting or harnessing NKp44–ligand interactions (for instance, using NKp44‐derived peptides that block inhibitory binding to PCNA) underscore the receptor’s promising potential as both a biomarker and a target in immunomodulatory interventions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "33", "end_ref": "39"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Claudia Cantoni, Marco Ponassi, Roberto Biassoni, et al.
