| HGNC Family | NLR family (NLR) |
| Name | nucleotide-binding oligomerization domain containing 2 |
| Description | This gene is a member of the Nod1/Apaf-1 family and encodes a protein with two caspase recruitment (CARD) domains and six leucine-rich repeats (LRRs). The protein is primarily expressed in the peripheral blood leukocytes. It plays a role in the immune response to intracellular bacterial lipopolysaccharides (LPS) by recognizing the muramyl dipeptide (MDP) derived from them and activating the NFKB protein. Mutations in this gene have been associated with Crohn disease and Blau syndrome. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nNOD2 is an intracellular pattern‐recognition receptor that plays a pivotal role in innate immunity by sensing specific bacterial cell wall components—most notably, muramyl dipeptide (MDP), a minimal bioactive motif derived from peptidoglycan. Upon binding MDP, wild‐type NOD2 undergoes conformational changes that trigger activation of nuclear factor (NF)-κB, thereby initiating pro‐inflammatory cytokine production and the expression of antimicrobial peptides. This activity is finely regulated by its carboxy-terminal leucine-rich repeat (LRR) domain and is disrupted by common Crohn’s disease–associated mutations (including frameshift and missense variants) that impair proper bacterial recognition and subsequent signaling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its canonical role in microbial sensing, further studies have extended the functional repertoire of NOD2. It is expressed not only in monocytes and dendritic cells but also in intestinal epithelial cells and specialized Paneth cells, where it contributes to mucosal homeostasis. NOD2 activation has been shown to engage autophagy by recruiting key autophagy proteins such as ATG16L1 at sites of bacterial entry, thereby enhancing pathogen clearance and facilitating antigen presentation. Moreover, evidence indicates that NOD2 may act as a cytosolic viral receptor—using the MAVS adaptor to activate interferon-regulatory factor 3 and induce interferon-β production—and that its ligand specificity is finely tuned to distinguish subtle structural variations in bacterial peptidoglycan fragments. These findings underline the significance of NOD2 in modulating both antibacterial and antiviral responses as well as in the genetic predisposition to granulomatous inflammatory diseases like Crohn’s and Blau syndromes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "27"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nClinical and mechanistic studies further elucidate that defects in NOD2 function—whether due to impaired membrane targeting, altered recruitment of signaling intermediates (such as RIP2 and XIAP), or diminished induction of antimicrobial peptides like defensins—can undermine the intestinal barrier and promote dysregulated inflammation, as observed in Crohn’s disease. Vitamin D signaling, for example, has been found to upregulate NOD2 expression, thereby enhancing its capacity to synergize with other mediators (such as IL-32) in amplifying antimicrobial and anti-inflammatory responses. Moreover, the interplay between NOD2 and autophagy-related proteins (e.g. ATG16L1) underscores a critical pathway for bacterial handling that is compromised by disease-associated mutations. These integrated mechanisms, along with observed population-specific differences in NOD2 variant prevalence and disease correlation, highlight a multifaceted role for NOD2 in maintaining gut immune homeostasis and in orchestrating responses to both bacterial and viral insults."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "28", "end_ref": "49"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "J P Hugot, M Chamaillard, H Zouali, et al.
