| Name | progestagen-associated endometrial protein |
| Description | This gene is a member of the kernel lipocalin superfamily whose members share relatively low sequence similarity but have highly conserved exon/intron structure and three-dimensional protein folding. Most lipocalins are clustered on the long arm of chromosome 9. The encoded glycoprotein has been previously referred to as pregnancy-associated endometrial alpha-2-globulin, placental protein 14, and glycodelin, but has been officially named progestagen-associated endometrial protein. Three distinct forms, with identical protein backbones but different glycosylation profiles, are found in amniotic fluid, follicular fluid and seminal plasma of the reproductive system. These glycoproteins have distinct and essential roles in regulating a uterine environment suitable for pregnancy and in the timing and occurrence of the appropriate sequence of events in the fertilization process. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Oct 2015] |

Its distinct glycoforms—such as glycodelin‐A, ‐S, ‐F, and ‐C—display tissue‐specific glycosylation that modulates critical sperm functions. For example, in the female reproductive tract, glycodelin‐A binds to receptors on the sperm (including FUT5) to inhibit premature acrosome reaction and zona pellucida binding, while seminal plasma glycodelin‐S maintains sperm in an uncapacitated state. Moreover, glycodelin is produced by granulosa cells in the ovary and subsequently modified by cumulus cells, establishing an optimal microenvironment for fertilization and early embryonic development."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its reproductive duties, glycodelin exhibits potent immunomodulatory functions. It modulates T‐cell responses—raising the threshold for activation through interactions with surface molecules including CD45 (and possibly CD7)—thereby biasing cytokine production toward a Th2 profile and inducing apoptosis in activated lymphocytes via mitochondrial pathways. Moreover, glycodelin can suppress B cell activation and alter cytokine secretion in monocytes, collectively fostering an immunosuppressive environment that is critical for maternal tolerance during pregnancy."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "14"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nGlycodelin is also a key mediator of endometrial differentiation and uterine receptivity. Its expression is induced by progesterone—and further enhanced under the influence of histone deacetylase inhibitors—driving the transformation of endometrial glandular cells into a secretory phenotype that supports implantation and placentation. Abnormal expression or glycosylation of glycodelin has been linked to reproductive pathologies such as polycystic ovary syndrome, endometriosis, recurrent implantation failure, and early pregnancy loss. Its presence on markers of endometrial receptivity (e.g. pinopodes) and its role in fine‐tuning angiogenic and cytokine responses further underscore its essential function in establishing fetomaternal immune tolerance."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "15", "end_ref": "30"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nEmerging evidence also points to roles for glycodelin in tumor biology and in non‐reproductive tissues.
