Description The protein encoded by this gene can function as a guanine nucleotide exchange factor (GEF) and may play a role in intracellular signaling and cytoskeleton dynamics at the Golgi apparatus. Polymorphisms in the region of this gene have been found to be associated with spinocerebellar ataxia in some study populations. Alternative splicing results in multiple transcript variants. In the nervous system, particularly within Purkinje cells, a single nucleotide substitution in the 5′ untranslated region (a C‑to‑T change) has been repeatedly associated with autosomal dominant cerebellar ataxias—such as SCA31 and other 16q‑linked ataxias—suggesting that even mutations outside the coding region can provoke pathogenic protein aggregation and neuronal dysfunction. Mechanistically, PLEKHG4 activates key Rho GTPases—including RhoA, Cdc42, and Rac1—to drive these malignant phenotypes, whereas its stability is negatively regulated by the ubiquitin‑conjugating enzyme UBE2O, which mediates its ubiquitination and degradation.
