HGNC Family | Zinc fingers |
Name | peroxisome proliferator-activated receptor alpha |
Description | Peroxisome proliferators include hypolipidemic drugs, herbicides, leukotriene antagonists, and plasticizers; this term arises because they induce an increase in the size and number of peroxisomes. Peroxisomes are subcellular organelles found in plants and animals that contain enzymes for respiration and for cholesterol and lipid metabolism. The action of peroxisome proliferators is thought to be mediated via specific receptors, called PPARs, which belong to the steroid hormone receptor superfamily. PPARs affect the expression of target genes involved in cell proliferation, cell differentiation and in immune and inflammation responses. Three closely related subtypes (alpha, beta/delta, and gamma) have been identified. This gene encodes the subtype PPAR-alpha, which is a nuclear transcription factor. Multiple alternatively spliced transcript variants have been described for this gene, although the full-length nature of only two has been determined. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nPPARα is a ligand‐activated nuclear receptor that functions as a master regulator of metabolic homeostasis. In the liver, heart, and skeletal muscle it senses endogenous fatty acids and related lipids to modulate the transcription of genes involved in fatty acid oxidation (e.g. carnitine palmitoyltransferase‐1A and pyruvate dehydrogenase kinase 4), lipoprotein metabolism (for example via apoAV induction), and even satiety signaling (through oleoylethanolamide production). Its transcriptional activity is finely tuned by several factors—including circadian regulators, bile acid signals via farnesoid X receptor, and even natural compounds such as the isoflavone genistein—to coordinate changes in energy expenditure and substrate use. Notably, alterations in PPARα expression or function have been linked to metabolic disorders such as nonalcoholic fatty liver disease, lipotoxic cardiomyopathy, and disruptions in fuel utilization; indeed, both endogenous signals (like fasting‐induced FGF21) and pharmacologic agents (for example, elafibranor) engage PPARα to restore insulin sensitivity, improve glucose homeostasis, and modulate inflammatory pathways."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its central role in metabolism, PPARα also governs key aspects of inflammation, cellular proliferation, and tissue adaptation. Structural studies have revealed that ligand binding influences PPARα’s interactions with co‐repressors (for example, SMRT) and co‐activators to control its transcriptional output. In immune cells, its activation dampens proinflammatory cytokine production and modulates the acute phase response. In vascular smooth muscle and even in certain breast cancer cells, PPARα can regulate cell‐cycle progression and proliferation. Furthermore, natural selection of specific PPARα haplotypes has been associated with high‐altitude hypoxia adaptation – a phenotype linked to decreased hemoglobin levels in Tibetan populations – while environmental chemicals (such as perfluoroalkyl acids and eicosanoids) have been shown to activate PPARα in a species‐dependent manner that may underlie differences in liver tumorigenesis. Finally, pharmacologic activation of PPARα holds therapeutic promise in a range of disorders, from cardiovascular disease and nonalcoholic steatohepatitis to neurodegenerative conditions via the stimulation of autophagy."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "31"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Deborah M Muoio, Paul S MacLean, David B Lang, et al.
