| Name | protein phosphatase 2 phosphatase activator |
| Description | Protein phosphatase 2A is one of the four major Ser/Thr phosphatases and is implicated in the negative control of cell growth and division. Protein phosphatase 2A holoenzymes are heterotrimeric proteins composed of a structural subunit A, a catalytic subunit C, and a regulatory subunit B. The regulatory subunit is encoded by a diverse set of genes that have been grouped into the B/PR55, B'/PR61, and B''/PR72 families. These different regulatory subunits confer distinct enzymatic specificities and intracellular localizations to the holozenzyme. The product of this gene belongs to the B' family. This gene encodes a specific phosphotyrosyl phosphatase activator of the dimeric form of protein phosphatase 2A. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nPTPA is an essential cellular regulator that activates protein phosphatase 2A (PP2A) by promoting a conformational change in its catalytic subunit. Its peptidyl‐prolyl cis/trans‐isomerase activity facilitates the proper folding and metal ion binding required for PP2A to achieve its pSer/Thr-specific phosphatase activity. In addition, PTPA is incorporated into a broader network of PP2A modulators that help prevent premature or promiscuous phosphatase activity until the full holoenzyme is properly assembled, thereby contributing to controlled cell proliferation and survival."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nStructural studies of PTPA reveal that it possesses a unique all α-helical fold organized into distinct subdomains (core, lid, and linker). A highly conserved surface patch mediates binding to PP2A, while a deep pocket accommodates ATP, enabling PTPA to participate in a composite ATPase activity when complexed with PP2A. This arrangement not only stabilizes the proper protein fold of the PP2A catalytic subunit but also positions ATP phosphoryl groups to modulate metal binding and ultimately alter the substrate specificity of PP2A. Mutations that impair PTPA’s cis/trans-isomerase or ATPase functions disrupt PP2A reactivation, underscoring its critical regulatory role."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jan Jordens, Veerle Janssens, Sari Longin, et al.
