| HGNC Family | Zinc fingers |
| Name | RAD18 E3 ubiquitin protein ligase |
| Description | The protein encoded by this gene is highly similar to S. cerevisiae DNA damage repair protein Rad18. Yeast Rad18 functions through its interaction with Rad6, which is an ubiquitin-conjugating enzyme required for post-replication repair of damaged DNA. Similar to its yeast counterpart, this protein is able to interact with the human homolog of yeast Rad6 protein through a conserved ring-finger motif. Mutation of this motif results in defective replication of UV-damaged DNA and hypersensitivity to multiple mutagens. [provided by RefSeq, Jul 2008] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRAD18 plays a central role in DNA damage tolerance by promoting trans‐lesion synthesis (TLS). In response to bulky DNA lesions and replication fork stalling, RAD18, in complex with RAD6, monoubiquitinates proliferating cell nuclear antigen (PCNA) to facilitate the recruitment and engagement of specialized low‐fidelity DNA polymerases such as Polkappa and Polymerase η. This post‐translational modification event is tightly regulated by upstream damage–sensing kinases and additional checkpoint signaling, thereby modulating polymerase switching and enabling error‐prone lesion bypass to ensure continued DNA replication even under genotoxic stress."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its function in TLS, RAD18 is fundamentally involved in homologous recombination (HR) repair and the activation of the Fanconi anemia (FA) pathway. It also modulates the chromatin recruitment and monoubiquitination of FA proteins (FANCD2 and FANCI), which is critical for the repair of interstrand cross‐links and for controlling the balance between HR and non‐homologous end joining (NHEJ) during replication stress."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "14"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nRegulation of RAD18 itself is multifaceted, involving self‐association, autoubiquitination, and defined structural domains (including its RING, SAP, and RAD6-binding domains) that determine its substrate specificity and complex formation. Its expression is further modulated by various factors—including microRNAs, competing endogenous RNAs (such as circRAD18), and interactions with cancer/testes antigens like MAGE-A4—as well as viral proteins that stabilize it.
