RAD9A Gene

Name RAD9 homolog A (S. pombe)
Description This gene product is highly similar to Schizosaccharomyces pombe rad9, a cell cycle checkpoint protein required for cell cycle arrest and DNA damage repair. This protein possesses 3' to 5' exonuclease activity, which may contribute to its role in sensing and repairing DNA damage. It forms a checkpoint protein complex with RAD1 and HUS1. This complex is recruited by checkpoint protein RAD17 to the sites of DNA damage, which is thought to be important for triggering the checkpoint-signaling cascade. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2011]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRAD9A is a critical component of the heterotrimeric 9‐1‐1 complex that orchestrates the cellular response to DNA damage. Acting as a “molecular sensor” at damaged or stalled replication forks, RAD9A is recruited through specific interactions—such as the binding of its phosphorylated C‐terminal tail with the basic cleft of the RPA70 OB‐fold—and its localization is tightly regulated by kinase‐mediated events (e.g. by PKCδ and Cdc2) to promote ATR–Chk1 signaling. RAD9A’s dynamic chromatin association supports multiple checkpoint functions including proper cell cycle arrest, telomere stabilization, and homologous recombination repair, with its rapid foci formation and cooperation with repair mediators like TopBP1 being essential for an effective response to genotoxic stress."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its checkpoint signaling role, RAD9A facilitates genome maintenance by directly modulating the activities of key DNA repair enzymes. Through physical interactions established by the 9‐1‐1 complex, RAD9A enhances the functions of repair proteins such as DNA polymerase β, FEN1, DNA ligase I, hNEIL1, and hOGG1—thereby promoting base excision and long‐patch repair pathways. Structural studies have identified unique interaction interfaces within the complex that distinguish RAD9A action from that of other sliding clamps like PCNA, and ubiquitylation‐dependent mechanisms (for example, via RAD18 binding to ubiquitylated chromatin) help recruit the 9‐1‐1 complex to DNA double–strand breaks. In a broader context, investigation of related signaling molecules (such as studies on T1AM distribution) further underscores the importance of tissue–specific factors in modulating these repair assemblies."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "25"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, RAD9A serves multifaceted regulatory functions that extend well beyond DNA damage sensing. As a transcriptional co–regulator, RAD9A modulates the activity of factors such as the androgen receptor and directly influences the expression of genes involved in cell survival, senescence and metastasis. Epigenetic deregulation of RAD9A, as observed in prostate cancers, along with regulation by kinases like TLK1 and its induction by bioactive compounds such as resveratrol, underscore its dual roles as either an oncogene or a tumor suppressor in different cellular contexts.
