| Name | ribosomal protein S6 |
| Description | Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a cytoplasmic ribosomal protein that is a component of the 40S subunit. The protein belongs to the S6E family of ribosomal proteins. It is the major substrate of protein kinases in the ribosome, with subsets of five C-terminal serine residues phosphorylated by different protein kinases. Phosphorylation is induced by a wide range of stimuli, including growth factors, tumor-promoting agents, and mitogens. Dephosphorylation occurs at growth arrest. The protein may contribute to the control of cell growth and proliferation through the selective translation of particular classes of mRNA. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. [provided by RefSeq, Jul 2008] |

RPS6 phosphorylation, mediated by kinases such as p70 S6 kinase, p90 ribosomal S6 kinase, and casein kinase 1, enhances the cap‐binding activity of the ribosome and modulates the efficiency of translation initiation. Recent ribosome footprinting analyses reveal that RPS6 becomes progressively dephosphorylated as ribosomes transit along mRNAs, a process that selectively promotes the translation of messages with short coding sequences. Moreover, RPS6 localization studies in primary versus transformed cells have further defined the compartmentalization of its phosphorylated versus non‐phosphorylated forms as a marker of active protein synthesis and a useful indicator of quiescent versus senescent states."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "4"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its canonical role in translation, RPS6 phosphorylation serves as a sensitive downstream readout of mTOR pathway activity in a range of physiological and pathological contexts. For example, in response to hormonal (leptin) signals, enhanced RPS6 phosphorylation in hypothalamic neurons regulates cap‐dependent translation to control energy homeostasis, while in skeletal muscle, resistance exercise robustly increases RPS6 phosphorylation to stimulate muscle protein synthesis. In cancer, hyperactivation of the mTOR signaling cascade is frequently reflected by elevated RPS6 phosphorylation, which has been employed as a predictive and prognostic biomarker in malignancies such as head and neck, esophageal, renal, and gastric cancers."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "5", "end_ref": "13"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its regulatory role in translation and growth signaling, RPS6 functions as a component of stress response assemblies. It has been detected as an integral molecule within stress granules in neuronal inclusions—suggesting a protective role against RNA oxidation in neurodegenerative settings such as Alzheimer’s disease—and modulates apoptotic responses; notably, the unphosphorylated form of RPS6 enhances TRAIL‐induced apoptosis by influencing death receptor expression. In addition, alterations in RPS6 phosphorylation have been linked to DNA damage responses and therapeutic resistance in BRCA1-deficient cancers, while RPS6 also interacts with viral proteins such as LANA to contribute to the stability of viral oncoprotein complexes."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "18"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFinally, a growing body of evidence indicates that RPS6 plays an important role in maintaining cancer stem cell phenotypes.
