RYR2 Gene

HGNC Family Ion channels, EF-hand domain containing
Name ryanodine receptor 2 (cardiac)
Description This gene encodes a ryanodine receptor found in cardiac muscle sarcoplasmic reticulum. The encoded protein is one of the components of a calcium channel, composed of a tetramer of the ryanodine receptor proteins and a tetramer of FK506 binding protein 1B proteins, that supplies calcium to cardiac muscle. Mutations in this gene are associated with stress-induced polymorphic ventricular tachycardia and arrhythmogenic right ventricular dysplasia. [provided by RefSeq, Jul 2008]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRyanodine receptor type 2 (RyR2) is a pivotal intracellular Ca²⁺ release channel localized to the sarcoplasmic/endoplasmic reticulum that underpins cardiac excitation–contraction coupling. It is dynamically regulated by post‐translational modifications, notably phosphorylation by protein kinases such as Ca²⁺/calmodulin-dependent protein kinase II and protein kinase A. Phosphorylation at discrete sites modulates RyR2’s Ca²⁺ sensitivity and open probability—mechanisms essential for augmenting contractility during adrenergic stimulation, yet potentially deleterious when aberrant, as observed in heart failure."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMutations in the RYR2 gene are closely linked to a broad spectrum of inherited arrhythmogenic disorders—including catecholaminergic polymorphic ventricular tachycardia (CPVT), exercise‐induced syncope, sudden cardiac death, atrial fibrillation, and even manifestations of cardiomyopathy. In these settings, missense mutations, deletions, or altered protein interactions (with regulatory partners such as calsequestrin, FKBP12.6, and calmodulin) disrupt normal channel gating. This disruption leads to abnormal Ca²⁺ release events (e.g., delayed afterdepolarizations and early afterdepolarizations) during adrenergic stress. Collectively, these studies underscore that both gain‐of‐function and, intriguingly, loss‐of‐function mutations can disturb Ca²⁺ homeostasis, while targeted pharmacological interventions (including flecainide and K201) and cascade genetic screening have helped to refine our understanding of RyR2‐related arrhythmogenesis and improve patient management."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "6", "end_ref": "34"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its central role in cardiac physiology, RyR2 influences Ca²⁺ signaling in diverse tissues, thereby affecting metabolic regulation and cell survival. For instance, in pancreatic β‑cells, altered RyR2 activity has been implicated in defective insulin secretion and in triggering apoptotic pathways—a mechanism that may contribute to the pathogenesis of type 2 diabetes mellitus. Moreover, emerging evidence highlights that RyR2 participates in non‐canonical signaling pathways in other cell types, including neurons and even melanoma cells, with findings that emphasize its complex regulation by alternative splicing, redox modifications, and dynamic interactions with accessory proteins (such as FKBP12.6) and partnering channels. Structural studies further delineate the intricate interaction between its cytoplasmic and transmembrane domains, collectively contributing to the precise modulation of intracellular Ca²⁺ homeostasis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "35", "end_ref": "37"}, {"type": "fg_f", "ref": "27"}, {"type": "fg_fs", "start_ref": "38", "end_ref": "44"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Xander H T Wehrens, Stephan E Lehnart, Steven R Reiken, et al.
