| HGNC Family | EF-hand domain containing, S100 calcium binding proteins (S100) |
| Name | S100 calcium binding protein A9 |
| Description | The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein may function in the inhibition of casein kinase and altered expression of this protein is associated with the disease cystic fibrosis. This antimicrobial protein exhibits antifungal and antibacterial activity. [provided by RefSeq, Nov 2014] |

At attomolar to nanomolar concentrations, S100A9 contributes to neutrophil chemotaxis and adhesion, modulating leukocyte trafficking and cytokine release in inflamed tissues. Through interactions with pattern‐recognition receptors such as the receptor for advanced glycation end‐products (RAGE) and Toll‐like receptors (TLRs), it initiates NF‐κB–dependent signaling cascades that lead to the production of proinflammatory mediators. The protein is highly expressed in neutrophils, monocytes and activated endothelial cells, and its extracellular release has been implicated in both local tissue inflammation (e.g. in rheumatoid arthritis and inflammatory bowel disease) and even in the pathophysiology of epithelial cancers where it contributes to proinflammatory tumor–stroma interactions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "14"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nOn a biochemical level, S100A9 is intricately regulated by divalent cations and undergoes calcium‐ and zinc‐dependent oligomerization that is essential for its biological activity. Moreover, S100A9 (and its heterocomplex with S100A8) binds key nutrients such as manganese and zinc, thereby mediating “nutritional immunity” by restricting access of these metals to invading pathogens. In addition, by binding fatty acids like arachidonic acid, it facilitates activation of NADPH oxidase and the production of reactive oxygen species, further contributing to antimicrobial defense. Its expression in various body fluids (e.g. fecal calprotectin in inflammatory bowel disease and amniotic fluid antimicrobial peptides) and on specific myeloid cell subsets (including myeloid‐derived suppressor cells and activated macrophages in juvenile idiopathic arthritis) underscores its utility as a biomarker of disease activity and tissue damage in disorders ranging from atherosclerosis and osteoarthritis to ulcerative colitis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "15", "end_ref": "30"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its roles in inflammation and host defense, S100A9 exerts multifaceted functions in tumor biology and related pathologies. In the tumor microenvironment, S100A9 produced both by myeloid cells and by tumor cells—with its engagement of receptors such as EMMPRIN and TLR4—promotes cell migration, invasion, and metastasis by activating p38 MAPK and NF‐κB signaling pathways. Paradoxically, S100A9 can also induce apoptotic and autophagy‐like cell death in neoplastic cells, underscoring its context‐dependent functions.
